RESUMO

Lactic acid bacteria (LAB) are the main microorganisms used as probiotics against gastrointestinal inflammation. The objective of this study was to evaluate the potential probiotic characteristics (antimicrobial activity, artificial gastrointestinal model resistance, cell surface hydrophobicity, and autoaggregation ability) and safety characteristics (hemolytic activity, antimicrobial resistance, and in vivo safety) of LAB isolated from the rumen fluid of an adult female sika deer. Two isolated strains identified as Latilactobacillus curvatus Z12 and Z19 showed good antimicrobial activity against enteropathogenic Escherichia coli (ATCC25922), Salmonella typhi (ATCC14028), and Staphylococcus aureus (ATCC25923). In addition, L. curvatus Z12 exhibited higher artificial gastrointestinal model resistance, cell surface hydrophobicity and autoaggregation ability than L. curvatus Z19. Therefore, regarding safety characteristics, only L. curvatus Z12 was evaluated. Upon assessment of safety, L. curvatus Z12 was negative for hemolytic activity and susceptible to penicillin G and cefamandole. Furthermore, an in vivo safety assessment showed that high-dose L. curvatus Z12 (109 CFU/mL) supplementation not only had no adverse effects on body weight gain, feed intake, and organ coefficients of treated mice but also played a key role in promoting the immune system maturation of treated mice. This research revealed that L. curvatus Z12 possesses desirable probiotic characteristics and could be used as a potential probiotic feed additive to improve sika deer health.

Based on a long-term simulated acid rain experiment, soil N2O emission fluxes were measured using static chambers and the gas chromatography method in a coniferous and broadleaved mixed forest and a monsoon evergreen broadleaved forest in southern China. During the five-year observation periods (2014-2018), soil N2O emission fluxes in the two forests showed obvious seasonal variation. The soil N2O emission fluxes in wet season were significantly higher than that in dry season, with a large annual variation. Due to the decreases of precipitation, soil N2O emission fluxes of the two forests in 2017 and 2018 were generally low. Soil N2O emission flux was positively correlated with soil temperature and soil moisture. In the monsoon evergreen broadleaved forest, soil N2O emission flux in the control plot was 12.6 µg N2O·m-2·h-1. Soil N2O emission fluxes under the pH 3.5 and pH 3.0 treatments increased by 42.9% and 61.1%, respectively. Soil N2O emission was significantly increased under simulated acid rain in the monsoon evergreen broadleaved forest. Acid rain promoted soil N2O emission in the coniferous and broadleaved mixed forest, but without significant difference among the treatments. Under the scenario of increasing acid rain, soil N2O emission fluxes in typical subtropical southern China forests would increase, and the magnitude of such increase was different among forest types.

AIMS/INTRODUCTION: To explore relationships between polyunsaturated fatty acids (PUFA) and non-alcoholic fatty liver disease (NAFLD) in patients with type 2 diabetes, and whether insulin action has an interactive effect with PUFA on NAFLD progression. MATERIALS AND METHODS: We extracted clinical and omics data of 482 type 2 diabetes patients from a tertiary hospital consecutively from April 2018 to April 2019. NAFLD was estimated by ultrasound at admission. Plasma fasting n3 and n6 fatty acids were quantified by liquid chromatography-tandem mass spectrometry analysis. Restricted cubic spline nested in binary logistic regression was used to select the cut-off point, and estimate odds ratios and 95% confidence intervals. Additive interactions of the n6 : n3 ratio with insulin action for NAFLD were estimated using relative excess risk due to interaction, attributable proportion due to interaction and synergy index. Relative excess risk due to interaction >0, attributable proportion due to interaction >0 or synergy index >1 indicates biological interaction. Spearman correlation analysis was used to obtain partial correlation coefficients between PUFA and hallmarks of NAFLD. RESULTS: Of 482 patients, 313 were with and 169 were without NAFLD. N3 ≥800 and n6 PUFA ≥8,100 µmol/L were independently associated with increased NAFLD risk; n6 : n3 ratio ≤10 was associated with NAFLD (odds ratio 1.80, 95% confidence interval 1.20-2.71), and the effect size was amplified by high C-peptide (odds ratio 8.89, 95% confidence interval 4.48-17.7) with significant interaction. The additive interaction of the n6 : n3 ratio and fasting insulin was not significant. CONCLUSION: Decreased n6 : n3 ratio was associated with increased NAFLD risk in type 2 diabetes patients, and the effect was only significant and amplified when there was the co-presence of high C-peptide.

BACKGROUND AND OBJECTIVE: Fufang Danshen formula, a famous Chinese patent medicine containing Salvia miltiorrhiza, Panax notoginseng and borneol, has been widely used in the treatment of coronary heart disease. The application is restricted by low bioavailability partly due to Panax notoginseng saponins (PNS) instability and low in vivo absorption. Thus, adhesive pellets were developed to improve bioavailability. The objectives of the present study were to evaluate the adhesive preparation by describing PNS's plasma pharmacokinetics in vivo and compare adhesive micro pills with normal preparation. METHOD: LC-MS/MS method was established to analyze five ingredients, notoginsenoside R1 (R1), ginsenoside Rg1 (Rg1), ginsenoside Rb1 (Rb1), ginsenoside Re (Re), and ginsenoside Rd (Rd), in rats' plasma to describe the pharmacokinetic parameters of PNS. RESULTS: The pharmacokinetic parameters were significantly different after oral administration three formulations. The results show adhesive formulations are superior to Fufang Danshen tablet (FDT); there are differences between the two adhesive, but not obvious. CONCLUSIONS: It was found that the modification with adhesive materials improved PNS bioavailability in Fufang Danshen formula. These findings provide a way for further in vivo evaluation of different formulations.

RESUMO

This study investigated the anticancer effect of Shenqi Fuzheng (SQFZ) injection combined with docetaxel on lung cancer cell lines of A549 and Lewis lung cancer (LLC). SQFZ injection alone cannot inhibit the vitality of lung cancer cells, but the antitumor activity of SQFZ combined with docetaxel was significantly higher than that using docetaxel alone.

The prescriptions including Polygoni Multiflori Caulis that built by Pro. Yan were collected to build a database based on traditional Chinese medicine (TCM) inheritance assist system. The method of association rules with apriori algorithm was used to achieve frequency of single medicine, frequency of drug combinations, association rules between drugs and core drug combinations. The datamining results indicated that in the prescriptions that including Polygoni Multiflori Caulis, the highest frequency used drugs were parched Ziziphi Spinosae Semen, Ostreae Concha, Ossis Mastodi Fossilia, Salviae Miltiorrhizae Radix Et Rhizoma, Paeoniae Rubra Radix, and so on. The most frequent drug combinations were "Polygoni Multiflori Caulis-parched Ziziphi Spinosae Semen", "Ostreae Concha-Polygoni Multiflori Caulis", and "Polygoni Multiflori Caulis-Ossis Mastodi Fossilia". The drug association rules of confidence coefficient 1 were "Ostreae Concha-->Polygoni Multiflori Caulis", "Poria-->Polygoni Multiflori Caulis", "parched Ziziphi Spinosae Semen-->Polygoni Multiflori Caulis", and "Paeoniae Alba Radix-->Polygoni Multiflori Caulis". The core drug combinations in the treatment of insomnia were Ossis Mastodi Fossilia, Polygoni Multiflori Caulis, Salviae Miltiorrhizae Radix et Rhizoma, Ostreae Concha, Polygalae Radix, Margaritifera Concha, Poria, and parched Ziziphi Spinosae Semen. And the core drug combinations in the treatment of obstruction of Qi in chest were Salviae Miltiorrhizae Radix Et Rhizoma, Polygoni Multiflori Caulis, parched Ziziphi Spinosae Semen, Trichosanthis Fructus, Allii Macrostemonis Bulbus, and Paeoniae Rubra Radix.
